Background: Abnormalities in mitosis parameter and cycle cell program are announced to be based on both the cell proliferation conditions and the condition of the metaphases of each person to respond to Phytohaemagglutinin (PHA) working condition. The aim of the current research is hence to comparison of mitotic index outcome in the individuals of normal volunteers involved in two different period of culture with effect of PHA.Methods: Comparison of healthy human donors lymphocytes were cultured for 48-h and 72-h and using conventional karyotype technique with minor modification. Results: Mitotic index and cell proliferation frequency were analyzed in the individuals of normal blood donors after preparing of colchcine to cultured metaphases 2 hours before cell fixation at. The metaphases indics at 48-h and 72-h culture period maintained a perfect yield level. Lymphocytes populations in 72-houres laboratory work were more than 48-h cultures (2.73% at 72 hours vs. 1.48% at 48 hours, p ≤ 0.05).Conclusion: The present research indicates significant comparisons for laboratories and individuals to be set up. With effect of these results it may help in time consuming and short time period for patient, as well as fresh suggestion for health care management decision. Reagent, manual, budget, time for diagnosis, and health care fulfillment may resolve of this proposal

Introduction: High resolution melting (HRM) analysis is a technique to measure the decrease of fluorescing, caused by a discharge of the dye, throughout the DNA thermal melting gradient process. STK11 gene encodes one of the cellular serine-threonine kinase proteins that regulate cellular polarity. It also acts as a tumor suppressor protein. Mutations of the germline in the gene are coincident with Peutz-Jeghers syndrome and the potential of developing a variety of neoplasia.Method: In an experimental study, the genome DNA of 56 patients with digestive system cancer was extracted. Afterward, nucleotide changes over the gene were examined using Real-time PCR and High resolution melting (HRM).Findings: Nucleotide screening using the HRM technique revealed two types of SNP in introns 6 & 7 in 10 patients. Four patients showed homozygous C/T nucleotide changes [cluster id/dsSNP/rs9282860] in intron 6 and six patients showed heterozygous C/G nucleotide changes [cluster id/dsSNP/rs2075607] in intron 7. A comparison of HRM results with sequencing results indicated 100% conformity. Conclusion: Although no mutation was observed in exon section of the gene, primary screening of the STK11 gene to diagnose unknown nucleotide changes of germline and somatic in patients with neoplasia using HRM was feasible, easy, and cost-effective.

Introduction: Common cold is the most common acute upper respiratory tract disease in children. The efficacy of vitamins and minerals for duration, symptoms and complications of common cold has been evaluated in several studies. This study was performed to determine the efficacy of zinc sulfate on common cold in children aged 1-5 years attending to Javaheri hospital in 2012-2013.Materials and Methods: In this clinical trial study, 112 children with common cold aged 1 to 5 years who were admitted at Javaheri hospital in 2012-2013 were enrolled. They were randomly assigned to receive either conventional treatment including Acetaminophen, Pediatric cold and nasal drop as control group or the aforementioned drugs plus zinc sulfate 1 mg/kg/day for 5 days. The duration of symptoms and rate of complications were compared between two groups. Results: The results revealed that the duration of disease was significantly shorter in case group (P = 0.033). Also the frequency of nasal discharge, sneezing, cough and sinusitis was significantly less in case group (P<0.05).Conclusion: In summary, it can be concluded that zinc sulfate is effective in reducing the symptoms, duration and complications of common cold in children aged from 1-5 years within 24 hours of the disease onset.

Introduction: Allergic rhinitis is a common problem in global health so that a quarter of the world population suffers from this disease. Also, the disease has a negative effect on the physical/psychological/ social performance of affected people and greatly affects the quality of life of patients. As a result, studies on the impact of current therapies on improving quality of life are important. Therefore, in this study, we examine the effect of treatment with a nasal spray of fluticasone and montelukast in the quality of life of patients. Methods: 195 patients with allergic rhinitis aged 15-60 years from both sexes who candidates for treatment with nasal fluticasone (50 µg once daily) and montelukast (10mg per night) were randomly selected to examine the quality of life and the relative factors. The patient’s quality of life was measured prior and two weeks after the treatment, using Juniper internationally validated the questionnaire. Results: In the current epidemiological study, women (59.5%) had a higher proportion of people with allergic rhinitis. Additionally, the highest prevalence of allergic rhinitis was seen among housewives (29.7%). The frequency of mild continuous allergic rhinitis (22.6%), intermittent (17.9%), and severe continuous type (15.9%) and intermittent (43.6%) was calculated. The results of this study showed that quality of life in 66.2% of patients was known to have made before any treatment intervention, and 33.8% had moderate-quality life defects. Two weeks after the start of treatment with fluticasone and montelukast, 172 (88.2%) had a mild quality of life defect and 23 (11.8%) had moderate to severe impairment in quality of life. Among the symptoms of nasal congestion, itching, and runny nose, frequent sneezing and pruritus, and runny nose, there was only a significant relationship between itching and runny nose and impaired quality of life after treatment. However, there was no association among the severity of the disease before treatment, age, sex, occupation, or asthma and quality of life after treatment (P> 0.05). In addition, there was a significant difference between sleep disorder before and after treatment with fluticasone and montelukast (P <0.001)Conclusion: The results of this study showed that two-weeks combination therapy with fluticasone and oral montelukast, significantly improved the quality of life and sleep disturbance. Additionally, there was a significant relationship between pruritus and runny nose and impaired quality of life after treatment. This could be due to the improvement of these symptoms after receiving the treatments.

Subdural hematoma (SDH) is a rare but important sequel of spinal anesthesia. We present a case of intracranial SDH in a lady developing headache after Cesarean section. A 34-years-old primigravida at 39 weeks of pregnancy was scheduled as an elective Cesarean section (c/s). She had no significant problems and also the pregnancy was done without any complications. The spinal anesthesia was given at L3-4 level by 25 G Quincke needle. Dural puncture was done well and was atraumatic. Two hours after delivery the headache had begun. That was on the frontal area with no other symptoms such as nausea and vomiting, confusion and loss of consciousness. She received Corticosteroid and then her headaches improved relatively but a few hours after discharge, she developed a frontotemporal headaches, that was became more and more and in about one month, she had headaches almost every day. After two months, she suddenly experienced much more severe headache with the left side paresthesia and numbness. She referred to a hospital. On the admission, the ophthalmological and neurological exams were done. Ophtalmological exams were normal. The brain MRI revealed a small SDH in the right frontal region without pressure effect. The results of her lab data were totally in normal range. We found that the spinal analgesia could be the only possibility of SDH. On the sequential follow up brain MRI, the SDH was completely resolved and her headaches were abolished as well. Conclusion: It is recommended when the patient has a persistent and prolonged headaches or a changed character one after doing lumbar puncture, SDH should be considered as a serious complication, just like the aforementioned case.

Conotoxins are neurotoxic peptides synthesized by marine cone snail. They have high potential as pharmaceutical agents, particularly as analgesics for the treatment of neuropathic pain. Well-defined ω-conotoxin family members are GVIA, GVIB, GVIC, MVIIA, MVIIC and MVIID. Some are blockers of the neuromuscular junction and they block calcium channels, it is reasonable to do large-scale synthesis derivative of Conotoxin for pharmaceutical purposes. Because of the short length of the peptides, their synthesis is not hard, but the modifications to enhance the potency have not been successful up to now. The well known ω- conotoxin family (1TT3, 1TTK, 1DW5, 1CNN, 1OMG, 1MVI, 1DW4, 1MVJ, 2KM9 and 1FEO) crystallographic data were obtained from PDB data bank. VMD 1.8.2 was used to draw and analyze the Ramachandran plot after they were being simulated at critical physiologic and pathologic human body temperatures with Monte carlo method in MM+ force filed. The most stable structure in terms of potential energy probability was 1TT3. Except 1DW4 and 1DW5 which did not show any structural changes in terms of gyration radius, there were significant differences between crystallographic data, analyzed physiologic and pathologic structures of other PDB codes. This functionality will lead us toward drug designing new blocker drugs of Neuromascular junction by biologically inspired design drug technology.
